4- 二甲氨基吡啶是近年來廣泛用于化學(xué)合成的新型高效催化劑,其結(jié)構(gòu)上供電子的二甲氨基與母環(huán)(吡啶環(huán))的共振,能強(qiáng)烈激活環(huán)上的氮原子進(jìn)行親核取代,顯著地催化高位阻、低反應(yīng)性的醇和胺的?;柞;?、磺酰化、碳酰化)反應(yīng),其活性約為吡啶的104-6倍。在有機(jī)合成、藥物合成、農(nóng)藥、醫(yī)藥、染料、香料、高分子化學(xué)、分析化學(xué)中的?;⑼榛?、醚化、酯化及酯交換等多種類型的反應(yīng)中有較高的催化能力,對(duì)提高收率有極其明顯的效果,此外還可以用作相轉(zhuǎn)移催化劑用于界面反應(yīng)。其優(yōu)點(diǎn)表現(xiàn)在:催化劑用量小,通常只需反應(yīng) 底物摩爾數(shù)的0.01-5%即可,反應(yīng)產(chǎn)生的酸可用有機(jī)堿或無機(jī)堿中和;反應(yīng)條件溫和,一般室溫下即可進(jìn)行反應(yīng),節(jié)約能源;溶媒選擇范圍廣,在極性、非極性有機(jī)溶劑均可進(jìn)行;反應(yīng)時(shí)間短,用吡啶長時(shí)間反應(yīng),而用DMAP則數(shù)分鐘即完成反應(yīng),因而大大提高了勞動(dòng)生產(chǎn)率;收率高,如用吡啶幾乎不反應(yīng)的空間位阻大的羥基化合物,使用DMAP收率可達(dá)80-90%,從而可提高反應(yīng)收率和產(chǎn)品質(zhì)量并簡化工藝;副反應(yīng)少,氣味小,三廢少;由于DMAP優(yōu)良的催化性能,被稱為“超級(jí)催化劑”,它已成為有機(jī)合成工作者最常用的催化劑之一。國內(nèi)化學(xué)制藥行業(yè)已成功將其應(yīng)用于乙(丙)酰螺旋霉素、青蒿素琥珀酸酯、他汀類降脂藥等原料藥的生產(chǎn)中,改善了工藝條件,并取得良好的經(jīng)濟(jì)和社會(huì)效益。此外,還廣泛地應(yīng)用于復(fù)雜天然產(chǎn)物的全(半)合成上,如:PRAVASTATIN全合成中的乙?;?,TUBERCIDIN全合成中的三氟乙酰化,MUGINEIC ACID全合成中的乙?;蚑ERPESTACIN全合成中的苯甲?;?,以及西地那非(VIAGAR)全合成中的酰化等,在農(nóng)藥生產(chǎn)上,在胺菊酯的合成上用DMAP提高了收率和產(chǎn)品純度,在異氰酸酯合成氨基甲酸酯、菊酰氯合成擬除蟲菊酯的反應(yīng)中也有明顯的催化活性。在磷?;磻?yīng)的有機(jī)磷合成中,作用相當(dāng)顯著。
